Direct measurements of Bragg-reflection phases in x-ray crystallography

چکیده
We present a reference-beam x-ray-diffraction technique applied to a good-quality protein crystal. The technique is based on a multiple-beam diffraction principle and employs a modified oscillation camera in a Bragg inclined geometry, which allows a simultaneous measurement of both magnitudes and triplet phases of a large number of Bragg reflections from the sample. This work demonstrates that the reference-beam method can be a simple practical solution to the phase problem in x-ray crystallography. @S0163-1829~99!02417-0#

Solving the Phase Problem Using Reference-Beam X-Ray Diffraction
A new method of obtaining Bragg reflection phases in an x-ray diffraction experiment is presented. It combines the phase-sensitive principles of multiple-beam diffraction and x-ray standing waves, and allows direct phase measurements of many multiple reflections simultaneously using a Bragg-inclined oscillating-crystal geometry. A modified-two-beam intensity function is devised to extract the p...
متن کاملExpanded Distorted-Wave Theory for Phase-Sensitive X-Ray Diffraction in Single Crystals
Diffraction of x rays is a widely used technique for studying structural information in condensed matter. One of the main advantages of this structural technique is that the diffracted intensities measured in experiments can be easily interpreted by a simple kinematic theory [1–3] that is based on the first-order Born approximation where single scattering events are the predominant mechanism. T...
متن کاملMapping the continuous reciprocal space intensity distribution of X-ray serial crystallography
Serial crystallography using X-ray free-electron lasers enables the collection of tens of thousands of measurements from an equal number of individual crystals, each of which can be smaller than 1 µm in size. This manuscript describes an alternative way of handling diffraction data recorded by serial femtosecond crystallography, by mapping the diffracted intensities into three-dimensional recip...

© 2006 American Institute of Physics, S-0031-9228-0603-030-4 M molecular biophysics is built on the twin pillars of genetic engineering and macromolecular structure determination. And x-ray crystallography is foremost among the methods used to determine the atomic-scale structure of macromolecules—very large molecules with sizes typically ranging from 50 to 1000 Å. X-ray crystallography played ...
متن کاملX-ray studies of the phases and phase transitions of liquid crystals.
A short review is given of recent X-ray diffraction studies of the phases and phase transitions of thermotropic liquid crystals. The areas covered are twist-grain-boundary phases, antiferroelectric phases studied with resonant X-ray diffraction and smectic phases within gel structures. In all areas, X-ray diffraction has played a key role. Nonetheless, open questions remain: the nature of the s...
